DNA Analysis Core Facility

Director: Johnson, Kyle, Ph.D.

Summary:

The DNA Analysis Core Facility supports researchers in their nucleic acid-based research studies on a 24/7 basis. The facility provides access to a wide range of genomic research tools including: DNA sequencing; microsatellite and fragment length polymorphism analysis; quantitative and real-time PCR; microarray analysis; and sophisticated nucleic acid analysis software. The DNA Analysis Core Facility services are available to external researchers from the RCMI program and other institutions for a nominal fee.

Resources:

Instruments

  • ABI 3130xl genetic analyzer ( DNA sequencer )

"16-capillary electrophoresis sequencer used for a variety of sequencing and fragment analysis applications including microsatellite analysis, AFLP, LOH,SNP validation, and SNP screening, as well as de novo sequencing and resequencing (mutational profiling)"

  • AKTA FPLC system ( Fast protein liquid chromatography instrument )

"Automated liquid chromatography system designed for research scale purification of proteins.
    It features:
Flow rates up to 20ml/min and pressures up to 5MPa
One working platform for all liquid chromatography techniques suitable for protein purification, from micro-gram to gram scale.
Method templates as a basis for creating customized methods
    Consists of:
    A compact separation unit
A personal computer running UNICORN software
    A fraction collector"

  • BioRobotics MicroGrid ( Microarray platform )

"The MicroGrid is designed for high throughput sample handling with the ability to work from 96, 384 or 1536 well microplates. Up to 24 microplates at a time can be accommodated in the BioBank loading cartridge and a maximum of 10 BioBanks (240 microplates) may be programmed into a single run.

The MicroGrid can be used to create everything from microarrays of DNA product on glass slides to macroarrays of bacterial colonies. Other accessories provide added functionality such as the management of bacterial libraries through replication and re-arraying."

  • GeneAmp PCR System 9700 ( Thermal cycler )

"GeneAmp PCR System 9700 is specifically designed for the amplification of nucleic acids. It is designed for use with 0.2 ml reaction tubes or 96-well reaction plates for all routine PCR applications"

Services

  • DNA Core Equipment Access ( Access service )

"Most of the Core equipment is accessible as a shared resource, operating on a 24-hour, 7-day per week basis, at no charge to the users. The Core provides equipment including thermocyclers for gradient PCR and qRT-PCR, storage PhosphorImager, UV/VIS and fluorescence spectrophotometers, SpeedVac concentrator, and digital gel documentation system.

Core personnel provide oversight and training in the proper use of the equipment."

  • DNA Sequencing Service ( Material analysis service )

"The DNA Analysis Core Facility also provides service on a recharge basis for DNA sequencing (on a full-service or ready-to-run basis)."

  • DNA/protein microarray printing ( Material processing service )

"The DNA Analysis Core Facility also provides service on a recharge basis for ... custom DNA/protein microarray printing."

Software

  • UNICORN ( Software )

"UNICORN is a complete package for control and supervision of chromatography systems. It consists of control software and a controller card for interfacing the controlling PC to the chromatography liquid handling module."
